مقایسه توانایی گره سازی جدایههای ریزوبیومی تودههای بومی عدس تحت تنش خشکی

<p>برای تعیین جدایههای متحمل به خشکی باکتریهای ریزوبیومی همزیست عدس، 12 نمونه خاک ازمناطق مختلف استانهای گلستان، چهار محال و بختیاری و اصفهان جمع آوری شد و ارقام محلی عدس بی نام دانه درشت، قزوینی و فریدنی در هر نمونه خاک در گلخانه کشت شد. پس از10 هفته از گرههای تشکیل شده روی ریشه گیاهان، 324 سویه ریزوبیومی جداسازی شدند. در تعیین تحمل به شوری جدایهها، مشخص شد که تمام جدایههای به دست آمده قدرت رشد در محیط کشت حاوی 200 میلیمولار کلرور سدیم را دارند. در مقادیر بالای نمک (بیش از 400 میلی مولار)، از نظر تحمل به شوری، تفاوت عمدهای در بین جدایهها وجود داشت، به طوری که فقط 20 درصد از آنها به عنوان متحمل به شوری ارزیابی گردید. جدایههای 249 RL و 211 RL با رشد در غلظت 550 و 600 میلی مولار نمک طعام به عنوان جدایههای برتر متحمل به شوری برگزیده شدند. نتایج بررسی تحمل به تنش پتانسیل ماتریک جدایهها در سطوح مختلف 6000 PEG با تحمل به شوری آنها مطابقت داشت. به طور کلی، جدایههای متحمل به شوری قادر به تحمل تنش خشکی در شرایط آزمایشگاهی نیز بودند، ولی این تحمل به شوری و خشکی ارتباطی با منشأ جغرافیایی این جدایهها نداشت. در یک طرح فاکتوریل، کرتهای خرد شده با سه تکرار، گرهسازی جدایههای متحمل 249 RL و 211 RL و نیز جدایه حساس 77 RL بر روی دو رقم عدس بی نام دانه درشت و فریدنی تحت تیمارهای مصرف 50، 75، 90 و 98 درصد آب قابل استفاده از خاک مقایسه شد. گرچه تنش خشکی به طور معنیداری باعث کاهش گرهسازی شد، اما رقم عدس بی نام دانه درشت به دلیل تراکم زیاد ریشه در واحد حجم، میزان گره سازی بیشتری داشت. با وجود این که، جدایه 249 RL در تحمل به شوری و خشکی در شرایط آزمایشگاهی و نیز آزمایشهای گلخانهای نسبت به سایر جدایهها برتری نشان داد، ولی با افزایش تنش خشکی در سطح بالاتر از 50 درصد مصرف آب قابل استفاده گرهزایی کاهش معنیداری داشت. </p>
To investigate drought tolerant isolates of rhizobial symbioant of lentil (<em>Lens culinaris</em> L.), 12 soil samples were collected from cultivated and non-cultivated area of Golestan, Chaharmahal-O-Bakhtiari and Isfahan provinces. Local cultivars of lentil including Binam Dorosht, Ghazvini and Faridani were planted in each soil sample. After 10 weeks, a total of 324 rhizobial isolates were recovered from root nodules of the lentil plants. Evaluation of the ability of the isolates to grow at different concentration of salt showed that all isolates grew normally on 200 mM NaCl and only 20% was determined as salt tolerant isolats(>400mM). Among the isolates RL249 was classified as superior salt tolerant strain due to growing on 600 mM salt. The drought tolerance of the isolates was also examined, using PEG6000. In general, the salt tolerant isolates were also drought tolerant, however their tolerance to salinity and drought is not related to their geographical origin. In a randomized split factorial design with three replications, the effectiveness of tolerant isolates(RL249 and RL211) and a sensitive strain (RL 77) was compared on two cultivars of lentil (Binam Dorosht and Faridani) under water stress treatments with the consumptions of 50, 75,90 and 98% of soil available water. Although nodulation rate was reduced in both cultivars as the consequence of drought stresses, plants of Binam Dorosht cultivar showed high nodulation rate due to the increased fresh weight of the roots. Despite the fact that RL249 was identified as a superior nodulating and salt/drought isolate, however nodulation efficiency was decreased significantly under water stress treatments with more than 50 % of soil available water.
