تأثیرکلرید سدیم بر فعالیت آنزیم لیپوکسی ژناز، میزان پراکسید هیدروژن پراکسیداسیون چربی در دانهالهای سه پایه پسته

یکی از عوامل مهم در تعیین مقاومت به کلرید سدیم، کارایی غشاهای سلولی در شرایط تنش میباشد. پراکسیداسیون چربیها ناشی از فعالیت آنزیم لیپوکسی ژناز و یا رادیکالهای آزاد اکسیژن، از عوامل مهم تخریب غشاهای سلولی در شرایط تنش کلرید سدیم است. در پژوهش حاضر آثار تیمارهای صفر، 75 و 150 میلی مول کلرید سدیم طی یک دوره چهارده روزه بر فعالیت آنزیم لیپوکسی ژناز، میزان پراکسید هیدروژن و مالون دی آلدهید ( پراکسیداسیون چربی) در برگ دانهالهای بنه (<em>Pistacia mutica</em> F.& M)، پسته قزوینی و پسته وحشی سرخس (<em>P. vera</em> L) بررسی گردید. نتایج نشان داد کلرید سدیم موجب افزایش فعالیت آنزیم لیپوکسی ژناز در برگ دانهالهای هر سه پایه پسته گردید و این فعالیت در روز هفتم پس از کاربرد تیمارهای کلرید سدیم به حداکثر رسید و در روز چهاردهم کاهش یافت. در دانهالهای بنه بیشترین میزان فعالیت آنزیم دیده شد و در روز چهاردهم کاهش کمتری نسبت به دانهالهای سرخس و قزوینی نشان داد. میزان پراکسید هیدروژن، در برگ دانهالهای هر سه پایه افزایش یافت و بیشترین میزان آن به ترتیب در دانهالهای بنه و سرخس پس از کاربرد تیمار کلرید سدیم 150میلیمول در روز چهاردهم مشاهده گردید. در دانهالهای هر سه پایه، میزان بالای مالون دی آلدهید (شاخص پراکسیداسیون چربی) در روزهای هفتم و چهاردهم دیده شد. نتایج هم چنین نشان داد امکان استفاده از شاخص پراکسیداسیون چربی و فعالیتهای مربوط به آن مانند آنزیم لیپوکسی ژناز در گزینش گیاهان مقاوم به کلرید سدیم در پسته وجود دارد و لازم است در مورد تغییرات بیوشیمیایی و آنزیمی در پسته در پاسخ به تنش کلرید سدیم بررسیهای بیشتری صورت گیرد.
One of the main criteria for salt tolerance is cell membrane stability under stress. Reactive oxygen species (ROS), activity of lipoxygenase (Lox) and lipid peroxidation are considered to be destructive to cell membrane under salt stress. In this study, the effects of 0, 75 and 150 mM NaCI, over a period of 14 days on the activity of lipoxygenease, the amount of hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>) and malon dialdehyde (MDA. as a lipid peroxidation) in the leaves of <em>Pistacia vera</em> L. (cv. 'Qazvini' and wild 'Sarakhs' pistachio) and 'Mastic' (<em>P. mutica</em> F. & M.) were studied. The results indicated that by increasing salinity Lox activity increased in the leaves of all the three rootstocks. This activity reached a climax on the 7<sup>th</sup> day and then decreased on the 14<sup>th</sup> day. Among the three species, Mastic reached the highest amount of the Lox activity on the 14<sup>th</sup> day with the lowest amount of Lox reduction. The amounts of H<sub>2</sub>O<sub>2</sub> in the leaves of all the three rootstocks increased, on the 14<sup>th</sup> day the highest amount of hydrogen peroxide was found in 'Mastic' and 'Sarakhs' after treating them with 150 mM NaCl. The amounts of MDA were also reached the highest level in all three rootstocks on the 7<sup>th</sup> and 14<sup>th</sup> days. The results also indicated the possibility of the use of lipid peroxidation index and Lox activity for selecting salt tolerant Pistacia rootstocks. More studies are needed for understanding the biochemical changes and enzyme activities in Pistacia rootstocks under salt stress.
